Store each kit component where it belongs
One box, two storage rules. Learn why the cultures go cold and dry while everything else stays on the shelf, and why getting it wrong shows up weeks later.
- Living material has storage conditions, chemicals mostly do not: A stain sits on a shelf for years. Dried cells are being kept alive but inactive, and warmth and moisture are what end that.
- Cold and dry are two separate requirements: A refrigerator handles the temperature. The desiccant in the packet handles the moisture. Meeting one of them is not meeting both.

- Step 1: Check each item against the table: Agar, stains and plates are all stored at room temperature by the protocol.
- Step 2: Find the one that is not: The cultures, which are the only entries with a temperature attached.
Why it's right: Three of the four are exactly where the protocol says they should live. Only the cultures carry storage conditions, so they are the only part of the box that a warm weekend can damage.
- A: The stains are stored at room temperature by the same table, so a bench is where they belong rather than a risk to them.
- B: Plates are stored at room temperature too, and a weekend indoors does not warp them.
- D: The nutrient agar is supplied as a bottle that is melted when it is needed, and room temperature is where the protocol keeps it.

- Step 1: Ask what you could see at the time: A vial that looks normal, because it does.
- Step 2: Ask when the truth arrives: Two days after a class streaks plates from it.
Why it's right: There is no signal at the moment of the mistake and no signal for weeks afterwards. The first evidence is a set of plates with nothing on them, by which time the cause is a month behind you.
- B: The box is opened to unpack it, so seeing inside is not the difficulty.
- C: That is true, and it explains why you cannot see the damage rather than why the mistake goes undetected for so long.
- D: Labels are not affected by room temperature, and a missing label would at least be something you could notice.

The four bacterial cultures go at 4 degrees Celsius with a . Everything else in the box is stored at temperature. A storage mistake here is invisible now and shows up as plates with no weeks later.
